We are seeking a highly motivated Cell Engineering Scientist/Senior Scientist to join our research team. The successful candidate will play a key role in building an industry-leading platform to develop life-saving allogeneic cell therapies. Essential Duties and Responsibilities

  1. Design genetic engineering strategies for next-gen allogeneic cell therapies.
  2. Generate reagents and develop protocols for engineering of primary immune cells and iPSCs.
  3. Maintain detailed records to support internal research and regulatory filing.
  4. Work closely with Immune Cell Therapy and iPSC Cell Therapy groups to advance R&D programs.

Skills and Competencies

  1. Must have hands-on experience with genetic engineering of primary immune cells and/or iPSC.
  2. Strong molecular skills to design and clone viral vectors and produce viral particles.
  3. Genomic biology skills to design and generate guide RNAs and targeting vectors.
  4. Experience in CRISPR On/Off-target analysis using PCR, qPCR, ddPCR and NGS.
  5. Lentivirus transduction and integration site analysis using LM-PCR and NGS.
  6. Bioinformatics skills including single cell RNA sequencing data analysis is desirable.
  7. Proficiency in designing, implementing, and troubleshooting flow cytometric experiments is preferred.
  8. Practical experience in iPSC single cell sorting, clonal expansion and characterization is preferred.
  9. Able to work independently as well as in a team with diverse background.
  10. Able to collect, analyze and present data to the research and management teams.
  11. Strong written and verbal communication skills. CRIPSR related publication/patent is desirable.
  12. A can-do attitude and an ability to adapt to evolving priorities. Able to travel when needed.

Educational Requirement

  1. PhD in Molecular Biology, Biochemistry, Immunology, Cell Biology, Bioengineering, or related field.

Overland Compensation and Benefits:

  1. The expected salary range for this position based on the South San Francisco is $125,000~$175,000. The pay ultimately offered will be determined based on job-related qualifications, education, experience, and other job related factors permitted by law.

We are seeking a highly motivated scientist (title and responsibilities to be adjusted as per experience) to join our research team. The successful candidate will play a key role in building an industry-leading cell therapy platform to develop life-saving cell therapies. Essential Duties and Responsibilities

  1. Establish NSG animal models for in vivo testing of CAR-T, CAR-NK and other cell therapy product candidates.
  2. Establish in vivo assays for functional analysis of CAR-T, CAR-NK cells.
  3. Establish and maintain cancer cell lines for in vitro and in vivo assays.
  4. Write and review in vivo studies protocols, schedules and coordinate with study directors.
  5. Manage vivarium operations and CRO relations, including equipment and consumables.
  6. Manage, and keep animal protocols updated for new cancer models and study procedures.
  7. Perform tumor burden measurements by volume and live animal imaging.
  8. Maintain organized, detailed records to support internal documentation and regulatory requirements.

Skills and Competencies

  1. Must have routinely performed xenograft cancer models using NSG or similar mouse strains.
  2. Must have live animal imaging system experience.
  3. Experience in administering CAR-NK or CAR- T cells to mouse models.
  4. Experience in monitoring animal health, GVHD, and tumor growth.
  5. Experience in isolating immune cells from tumor and tissues.
  6. Knowledge and experience with allogeneic CAR-NK or CAR-T is preferred.
  7. Able to work independently as well as in a team with diverse background.
  8. Able to collect, analyze and present data to the research and management teams.
  9. Strong written and verbal communication skills, and detailed record keeping skills.
  10. A can-do attitude and an ability to adapt to evolving priorities.

Educational Requirement

  1. PhD., MS. or BS. in Immunology, Bioengineering, Biology, Animal Sciences, or related fields with over 4 years of cell therapy in vivo model related academic or industry experience.

Essential Duties and Responsibilities

  1. Drive the generation of high-quality biologics to advance the cell therapeutics pipeline by leading as well as independently performing antibody discovery campaigns using in vitro antibody discovery and optimization techniques.
  2. Apply multiple cutting-edge selection techniques to synthetic and immune antibody libraries, like phage display, yeast display, FACS sorting, plate-based screening.
  3. Use automation to support antibody production, purification, and screening.
  4. Sub-clone hits into expression vectors and produces them at micro-scale in a high-throughput manner.
  5. Characterize binders’ properties in various affinity and activity assays.
  6. Engineer and improve binders by means of affinity maturation and/or humanization.
  7. Independently generate hypotheses, troubleshoot issues, design and execute experiments.
  8. Supervise CRO activities.
  9. Document the results in a detailed manner in Laboratory journal and in internal Databases.
  10. Present research updates and participate constructively in working project teams and at laboratory group meetings.
  11. Support business development scientific due diligence as subject matter expert.

Skills and Competencies

  1. At least 5+ years of experience in antibody discovery technologies, experience with binder discovery and CAR design for cell therapy is preferred.
  2. Knowledge in molecular biology, cell biology, biochemistry, affinity, and activity assays.
  3. Experience in antibody discovery through naive or immune library panning as well as engineering.
  4. Experience in mammalian and bacterial protein production encompassing expression and purification.
  5. Experience in flow cytometry and lab automation.
  6. Experience in bispecific antibody or immune cell engager design and discovery is preferred.
  7. Knowledge in NGS methodologies.
  8. Familiarity with structural bioinformatics.
  9. Experience in working effectively in matrix environment and collaboration with different functional areas.
  10. Familiarity with structure/function relationship and structural interpretation.
  11. Experience in project management/leadership.

Educational Requirement

  3. Experience in antibody discovery technologies and CAR-T binder discovery and engineering is strongly preferred.

Essential Duties and Responsibilities

  1. Serve as internal immunology expert to develop allogeneic immune cell therapies.
  2. Develop flow assays for phenotypic analysis of iPSC-derived NK and T cells.
  3. Develop target cell killing assays for functional analysis of iPSC-derived NK and T cells.
  4. Develop benchmark assays using primary NK and T cells.
  5. Develop reagents and assays to support CAR binder generation and characterization.
  6. Maintain organized, detailed records to support internal documentation and regulatory requirements.
  7. Manage lab operations, such as equipment maintenance and ordering of consumables.

Skills and Competencies

  1. Must have hands-on experience with human immune cell isolation, phenotypic and functional analysis.
  2. Experience with immune cells ex vivo activation, viral transduction and expansion is required.
  3. Ex vivo T or NK cell activity analysis is required. Experience with Incucyte is preferred.
  4. Experience in ELISA or similar assays to measure cytokines is required.
  5. Experience with CAR-NK or CAR-T In vivo efficacy models using NSG mice is preferred.
  6. Knowledge and experience with allogeneic CAR-NK or CAR-T is preferred.
  7. Knowledge and experience with gamma-delta T cells is desirable.

  9. Experience in iPSC differentiation and immune cell therapy under cGMP condition is preferred.
